Hettich EBA 12 Refrigerated Benchtop Centrifuge Used in Semi-Industrial Scale Alginate Production Study
The Hettich EBA 12 refrigerated benchtop centrifuge was utilized in the study “Production of alginate by Azotobacter vinelandii in semi-industrial scale using batch and fed-batch cultivation systems,” published in the Journal of Advanced Scientific Research. The study aimed to produce alginate using Azotobacter vinelandii bacteria in batch and fed-batch cultivation processes on a semi-industrial scale level with the help of 16-L and 150-L stirred tank bioreactors (STR).
According to the study, “the importance of alginates is based on their wide industrial applications as thickening agents, stabilizers, gelling agents, and emulsifiers in food, textile, and paper making industries.” The Hettich EBA 12 centrifuge was used for the study’s sample preparation technique. 10 mL broth samples were centrifuged using the Hettich EBA 12 at 6000 rpm and 20 °C for 40 minutes after having 0.2 ml of Na4EDTA (0.5 M) and 0.2 ml of NaCl (5m) added to the broth samples.
The researchers involved in the study used a controlled pH culture and uncontrolled pH culture to determine the batch cultivation’s generation of the maximal volumetric alginate production and maximal specific alginate production. Once glucose was detected as the limiting substrate of the process, fed-batch cultivation was conducted with the use of a C-source constant feeding strategy and other medium components for increasing the production of volumetric alginate.
The results of the study indicated that the fed-batch cultivation strategy displayed a high potential for large scale alginate production and almost reached the maximal theoretical alginate yield.
