The Hettich ROTANTA 460 benchtop centrifuge was used for processing blood samples during research in the collaborative study entitled “Experimental infection of chickens with Histomonas meleagridis confirms the presence of antibodies in different parts of the intestine” by M. Windisch and M. Hess from the University of Veterinary Medicine and Department for Farm Animals and Veterinary Public Health in Vienna. The researchers sought “to determine the intestinal mucosal immune response in specific pathogen-free chickens.”

Specific pathogen-free (spf) chickens were infected with a clonal culture of the protozoan parasite Histomonas meleagridis. Samples of birds were euthanized at regular intervals during the study, starting the day before infection. Euthanized birds were necropsied and tissue samples were taken from the caeca, duodena and jejuna regions of the intestines.

At each sampling date, blood samples were also taken from all birds in the study. The blood samples were centrifuged using the Hettich ROTANTA 460 and the acquired serum was stored at -20°C for later use.

Ex vivo cultures were produced from the intestinal samples to determine antibody levels. “An indirect sandwich ELISA was used as described recently to measure the presence of [antibodies] IgG, IgA and IgM in both, serum samples and the supernatants of the ex vivo cultures.”

“Generally,” the study says, “an immunological reaction was noticed in all parts of the gut.” “In the sera, a continuous increase of IgG antibodies was found with the first positive samples above the cut-off value at 14 d.p.i.” “In comparison, serological levels for IgA and IgM remained lower with a clear peak at 28 days p.i.”
