Hettich ROTOFIX 32 Used to Develop Preservative-Free TACA Suspension
A study entitled “Preservative-free Triamcinolone Acetonide Suspension Developed for Intravitreal Injection” was published in the Journal of Ocular Pharmacology and Therapeutics in 2008. It detailed the development of a reproducible method involving Hettich’s ROTOFIX 32 centrifuge for compounding preservative-free TACA suspensions.
According to the study, triamcinolone acetonide (TACA) therapy is commonly used to treat several retinal diseases. Patients suffering from ailments such as diffuse diabetic macular edema, exudative age-related macular degeneration, uveitis, and macular edema from central retinal vein occlusion can opt to undergo off-label therapy that requires TACA to be administered via intravitreal injection. Despite the promising results however, all commercially available TACA suspensions used for treatment contain retinal toxic vehicles. To remedy this, Christopher Bitter and his team of researchers aimed to find a convenient and reproducible method to compound a totally preservative-free TACA suspension with consistent quality.
Three methods for compounding TACA suspensions for intravitreal injection were evaluated in the study. These methods included membrane-filter, centrifugation, and direct suspending. The Hettich ROTOFIX 32 was used to centrifuge Kenacort A 40 samples for 2 minutes at 4000 rpm and the supernatant vehicle was rejected. TACA particles were washed two times with aliquots of Ringer’s solution Hartmann by vortexing, centrifugation, and rejection of the supernatant. Ultimately, the direct suspending method proved to be the most convenient, providing a TACA suspension that fulfilled the researcher’s requirements and a 2-year shelf life.
